⚡️ Allosteric JH2 Pseudokinase Locking Mechanism

Instead of competing for the conserved ATP-binding pocket, Zasocitinib (TAK-279) binds exclusively to the regulatory JH2 pseudokinase domain of tyrosine kinase 2. This mechanically locks the enzyme in an inactive state without altering catalytic pockets.

Selectivity for TYK2 is 170-fold higher than for JAK1 or JAK3. Consequently, pathogenic IL-23 and IL-12 inflammatory signaling is extinguished, while essential JAK2 hematopoiesis continues unaffected, eliminating cytopenia risks.
